I am trying to set up a 5 axis drilling program on a VMC and need some formulae for computing the corrections needed to account for fixture set up error.

Is there a way I can account for the fixture deviation in MasterCAM?

Draw it up from some known "Hard reference". Mastercam will always know where you are at. Just set the hard reference at system origin as if it were a five axis machine.

Nice start, bring on the details please.

DCAPPS,

Create a point at the orign of your MC file.

Make a copy of the top CPlane and make it associative to the point.

Build all your tool paths using the copied Top Cplane as your WCS.

Once you know how much your fixture is off, translate the point by that amount and regen.

Your toolpaths are tied to the point through the copied WCS. They will not move in Mastercam, but the posted gcode will.

If the post works OK, all you should have to do is select CPLANE, ALT-O, and select the point that's at the center of rotation. You don't have to translate the part or change WCS.
